Towards a Mechanistic Understanding of Kinase-Catalysed N-Phosphorylations Using Multiscale Simulations

Abstract:

Phosphorylierungsreaktionen sind zentrale Prozesse in biologischen Systemen. Obwohl seltener als Phosphorylierungen an Sauerstoffatomen, spielen Phosphorylierungen an Stickstoffzentren eine wichtige biologische Rolle. Beispiele hierfür sind die Autophosphorylierung von Histidin in Signaltransduktionskaskaden sowie die reversible Phosphorylierung von Kreatin zu Phosphokreatin, welches als kurzfristig verfügbare Energiereserve dient. Trotz ihrer biologischen Bedeutung sind die molekularen Mechanismen kinasekatalysierter N-Phosphorylierungen bislang nicht vollständig aufgeklärt.

Abstract (englisch):

Phosphorylation reactions are fundamental in biological systems. Although rarer than phosphorylation of oxygen atoms, nitrogen phosphorylations play important biological roles, including histidine autophosphorylation in signal transduction, and reversible phosphorylation of creatine to phosphocreatine, a rapidly mobilisable energy reserve. Despite their relevance, the detailed understanding of the molecular mechanisms of kinase-catalysed N-phosphorylation reactions remains incomplete.
Hybrid QM/MM methods enable the description of enzymatic processes at atomic resolution by employing quantum mechanical (QM) methods for the active site while treating the surrounding protein environment classically. ... mehr
